Abstract

Tanaman air adalah bagian penting dari ekosistem air tawar. Salah satu spesies yang terkenal adalah Bacopa australis. Hobiis aquascape saat ini memiliki ketertarikan tinggi terhadap tanaman air dengan kualitas yang bagus dari setiap spesiesnya. Metode perbanyakan tanaman air tanpa tanah, lahan pertanian dan air perlu dilakukan untuk memenuhi keinginan tersebut. Tujuan dari penelitian ini adalah untuk mendapatkan formula media kultur jaringan dan zat pengatur tumbuh yang tepat untuk multiplikasi dalam perakitan mother plant (tanaman induk) Bacopa australis, serta mendapatkan media terbaik untuk aklimatisasi. Media yang digunakan adalah media Murashige dan Skoog (MS) A padat dengan perbedaan konsentrasi zat pengatur tumbuh. Perlakuan uji dalam kombinasi zat pengatur tumbuh (a) 0,50 mg/L BAP + 0,50 mg/L kinetin; (b) 0,50 mg/L BAP; dan (c) 0,50 mg/L 2,4-D. Aklimatisasi tanaman induk dilakukan pada berbagai media antara lain 1) pasir silika + pupuk aqua soil amazonia, 2). pasir malang + pupuk aqua soil amazonia, 3) pasir silika + pupuk cair; 4) pasir malang + pupuk. Hasil yang diperoleh, yaitu formula media kultur terbaik untuk multiplikasi tunas tanaman B. australis secara in-vitro adalah media MS (A) yang diperkaya dengan 0,5 mg/L BAP + 0,5 mg/L kinetin, sedangkan aklimatisasi terbaik pada media pasir malang + pupuk aqua soil amazonia.Water plant is an important part of freshwater ecosystems. One of the famous species is Bacopa australis. Today, many aquascape hobbyists have a high interest in aquatic plant species that have good aesthetic appearances. To answer this challenge, a new method in-vitro propagation of aquatic plants, planted without soil, agricultural land and water was conducted. The aim of this research was to find the best growth regulator hormon formula and aclimatisation medium, in creating the mother plant Bacopa australis. The medium used was MS (Murashige and Skoog, 1974) with different growth regulator hormon, i.e: (a) 0.50 mg L-1 BAP + 0.50 mg L-1 kinetin, (b) 0.50 mg L-1 BAP, (c) 0.50 mg L-1 2.4-D. The aclimatisation of the mother plant candidates used four treatments, i.e: (1) silica sand + aqua soil amazonia fertilizer, (2) malang sand + aqua soil amazonia fertilizer, (3) silica sand + liquid fertilizer, (4) malang sand + liquid fertilizer. The results showed that the best formula for in-vitro multiplication mother plant of Bacopa australis was MS medium with 0.5 mg/L BAP + 0.5 mg/L kinetin (treatment A). The best medium aclimatisation was malang sand + aqua soil amazonia fertilizer medium.

Abstract

Pengolahan air buangan tambak superintensif (TSI) adalah usaha untuk mengurangi beban bahan pencemar yang terkandung di dalam air buangan TSI sehingga aman dan tidak membahayakan saat dibuang ke lingkungan. Penelitian ini bertujuan untuk mengevaluasi desain dan performansi Instalasi Pengolahan Air Limbah (IPAL) dalam memperbaiki kualitas air buangan TSI sebelum dibuang ke badan air. IPAL terdiri atas kolam sedimentasi, dua kolam aerasi, dan satu kolam penampungan. Ke dalam kolam penampungan ditebari ikan mujair serta rumput laut Gracilaria sp. yang dibudidayakan dengan metode long line, berfungsi sebagai biokontrol. Sampel air diambil di bagian inlet IPAL, oulet kolam sedimentasi atau inlet kolam aerasi-1, outlet kolam aerasi-1 atau inlet kolam aerasi-2, outlet kolam aerasi-2 atau inlet kolam penampungan, serta outlet kolam penampungan, setiap dua minggu selama 105 hari pemeliharaan. Parameter yang diukur adalah total padatan tersuspensi (TSS), total amonia nitrogen (TAN), nitrit, nitrat, fosfat, bahan organik terlarut (BOT), dan biological oxygen demand (BOD-5). Spesifikasi teknis IPAL yang diamati meliputi ukuran dan volume IPAL, volume dan waktu tinggal air buangan tambak, dan efisiensi kinerja IPAL, serta rasio volume IPAL dan volume total air tambak. Hasil penelitian menunjukkan bahwa IPAL dapat mengurangi beban bahan pencemar dengan tingkat efisiensi antara 53,1%-99,4%; namun masih diperlukan peningkatan kapasitas dalam mengurangi konsentrasi BOT. IPAL menghasilkan efisiensi yang tinggi terhadap TSS, TAN, nitrit, Total Nitrogen (TN), dan fosfat. Rasio volume IPAL dan volume air tambak 30:70 dengan waktu tinggal minimal lima hari, dapat dijadikan acuan dalam pembangunan IPAL tambak superintensif.A wastewater treatment plant (WTP) in a super-intensive shrimp farm is used to reduce organic matters contained in super-intensive shrimp farm effluent. Through the WTP, the waste water from shrimp facilities can safely and harmlessly be released to the receiving environments. The aims of this study were to evaluate the design and performance of a WTP in reconditioning waste water released from a super-intensive shrimp farm prior to release to water bodies. The WTP was made of a series of sedimentation pond, two aeration ponds, and one reservoir or equalitation pond. The tilapia fish and seaweed, Gracilaria sp., were stocked in the equalitation pond where the seaweed was cultured using long line method; these organisms were used as bio-control. Water samples were collected fortnightly during 105 days of culturing duration from the WTP inlet, outlet of sedimentation pond or at inlet of the first aeration pond; outlet of the first aeration pond or inlet of the second aeration pond, outlet of the second aeration pond or inlet of equalitation pond and the outlet of equalitation pond. The measured variables were total suspended solid (TSS), total ammonia nitrogen (TAN), nitrite, nitrate, phosphate, total organic matters (TOM), and five days biological oxygen demand (BOD5). The evaluated technical performances of the plant were its size and volume; volume and retention time of effluent, efficiency of WTP performance and volume ratios of the WTP and total volume of shrimp pond. The results of the study indicated that the WTP was able to reduce concentrations of nutrients and solids in effluent by 53.1%-99.4% of efficiency. However, its capacity need to be increased due to reducing concentrations of TOM. The WTP was highly efficient in reducing the concentrations of TSS, TAN, nitrite, total N, and phosphate. The volume ratios between the plant and pond waters were 30:70 with minimum retention time five which days could be proposed for wastewater treatment pond for super-intensive shrimp ponds.

Abstract

Tujuan penelitian ini adalah untuk mengaplikasikan pemanfaatan bahan baku lokal di Klungkung, Bali sebagai pakan ikan nila BEST dibandingkan dengan pakan komersil. Pemilihan bahan baku lokal untuk memformulasikan pakan uji ditentukan berdasarkan hasil survei bahan baku yang mengandung nutrien terbaik sesuai dengan kebutuhan ikan nila dan harga yang relatif murah. Pakan uji yang diformulasikan mengandung protein 29%-30%. Performa pakan uji dibandingkan dengan pakan komersil dengan menggunakan uji-T. Setelah diaklimatisasi, benih ikan nila BEST (panjang total 5,7 ± 0,4 cm; bobot 3,1 ± 1,8 g) ditebar secara acak ke dalam enam buah hapa berukuran 2 m × 1 m × 1 m yang terletak di dalam kolam 100 m2 dengan kepadatan 65 ekor/m3 dan diberi pakan uji sebanyak 3% dari total bobot seluruh ikan uji per hari selama tiga bulan masa pemeliharaan. Hasil penelitian menunjukkan bahwa pertumbuhan ikan nila BEST tidak berbeda nyata antara pakan uji hasil formulasi dengan pakan komersil (P>0,05). Nilai FCR pakan komersil tidak berbeda nyata dibandingkan pakan formulasi. Berdasarkan hasil tersebut, keuntungan dengan menggunakan bahan baku lokal lebih tinggi dibandingkan dengan pakan komersil.The purpose of this study was to compare the utilization effects of locally found feed ingredients in Klungkung, Bali for BEST strain tilapia feed compared to a commercial feed. The selection of local raw ingredients used in the feed formulation was based on the best nutrient composition for tilapia needs and its price determined through a separate survey. Formulated test feed contained 29%-30% protein. The formulated feed performance was compared to the commercial feed using the T-test. After acclimatization, the fish (BEST strain tilapia, total length of 5.7 ± 0.4 cm; weight of 3.1 ± 18 g) were randomly stocked into six hapas, each measuring 2 m × 1 m × 1 m in size placed inside a pond (100 m2) with a density of 65 fish/m3 and fed 3% of the total fish biomass per day for three months rearing period. The results showed that the growth of BEST tilapia was not significantly different between the formulated feed and commercial feed (P>0.05). The FCR value of the commercial feed was not significantly different compared to the formulation feed. Based on these results, the benefit of using local raw materials was higher than that of commercial feed.

Abstract

Isu nasional menurunnya produksi budidaya ikan bandeng di tambak pantai utara Pulau Jawa didugasebagai akibat rendahnya kualitas benih produk Hatchery Skala Rumah Tangga (HSRT) di Bali, yang secara kontinu merupakan sumber utama pasok benih. Banyak faktor yang mempengaruhi kualitas benih, antara lain kualitas telur dan induk, serta manajemen pemeliharaan induk dan larva. Tujuan penelitian ini adalah untuk mengevaluasi performa pertumbuhan,aktivitas enzim pencernaan dan nutrisi benih ikan bandeng dari HSRT dan generasi kedua (G2) terseleksi yang dipelihara berdasarkan standar operasional prosedur. Penelitian dilakukan di tambak Pejarakan, dengan hewan uji benih produk HSRT dan benih generasi ke-2 (G-2) terseleksi dengan panjang total rata-rata 11,79 ± 1,64 mm, masing-masing dengan padat tebar 5.000 ekor/petak dengan luasan 0,5 Ha/petak, diberi pakan jenis pelet kering berkadar protein 25 % dan dipelihara selama 6 bulan. Hasil penelitian menunjukkan performa benih ikan bandeng dipengaruhi oleh sumber induk dan manajemen pemeliharaan saat larva. Pertumbuhan benih ikan bandeng asal HSRT dengan SOP pemeliharaan larva menunjukkan peningkatan laju pertumbuhan panjang dan bobot sebesar 10,11% dan 47,18% lebih tinggi dibandingkan benih G2-terseleksi, dan 13,82% dan 50,55% lebih tinggi dibandingkan benih HSRT tanpa SOP. Aktivitas enzimatis pada benih HSRT dengan SOP lebih efisien dibandingkan benih G2-terseleksi. Aktivitas enzimatis pada benih HSRT tanpa SOP adalah yang paling rendah dimana hal ini terlihat dari laju pertumbuhannya yang juga paling rendah. Benih HSRT yang dipelihara dengan SOP mampu menekan rasio konversi pakan sebesar 28,29% lebih rendah dibandingkan benih G2-terseleksi, dan 22,64% dibandingkan benih HSRT yang dipelihara tanpa SOP. Currently, there is a national concern regarding the decreasing of milkfish production from ponds in North Java allegedly due to a low quality of milkfish seed produced by small-scale hatcheries in Bali, which is the main producer of milkfish seed. Some factors can influence seed quality, such as quality of egg and broodstock also rearing management of broodstock and larvae. The aim of this experiment was to evaluate morphological aspect (growth rate) and biological aspect (digestive enzymes activities) of seed from backyard hatchery and selected G2 milkfish. Research on grow-out of milkfish seed was conducted at the IMRAD ponds facility in Pejarakan, using milkfish seed produced by small-scale hatcheries as well as selected second-generation (G-2) seed, each with the density of 5,000 seed/pond (1 pond=0.5 ha). The seeds were fed with dry pellet and reared for 6 months.The result showed performance of seed in terms of morphological and biological influenced by broodstock itself and larvae rearing management.The growth of seed of HSRT origin with larvae rearing SOP had increased the length of and weight growth rates of 10.11% and 47.18%, respectively compared to seed G2 selected and 13.82% and 50.55% from seed HSRT without SOP. Enzymatic activity in HSRT seed with SOP was more efficient than that of selected G2 seed. Enzymatic activity in HSRT seed without SOP was the lowest in which correlated to the lowest growth. Seed from HSRT origin with SOP had better feed conversion ratio which was 28.29% lower than that of selected G2 seed and 22.64% lower than that of HSRT seed without SOP.

Abstract

Ikan uceng (Nemacheilus fasciatus) merupakan salah satu spesies ikan air tawar di Indonesia dengan nilai ekonomi cukup tinggi yang ketersediaannya masih mengandalkan penangkapan di alam, sehingga diperlukan upaya domestikasi untuk menjaga kelestariannya. Tujuan dari kegiatan ini adalah untuk mengamati biologi reproduksi dan pola pertumbuhan ikan uceng di lingkungan buatan (akuarium). Ikan uceng hasil tangkapan alam dari Sungai Progo, Temanggung, Jawa Tengah (panjang total 5,55 ± 0,53 cm; bobot 2,49 ± 0,24 g) diadaptasikan selama 12 bulan di akuarium (40 cm × 30 cm × 30 cm) dengan sistem air mengalir yang dilengkapi dengan aerator. Ikan uceng diberi Tubifex, hingga sampai akhirnya dapat beradaptasi dengan pakan komersial. Pakan komersial yang diberikan yaitu sebesar 3% per hari dari biomassa tubuh dengan frekuensi dua kali sehari. Data biologi reproduksi diperoleh melalui koleksi data panjang total, bobot badan, bobot gonad, fekunditas, diameter telur, dan indeks kematangan gonad. Data pola pertumbuhan diperoleh dengan koleksi data panjang, bobot, dan sintasannya. Hasil penelitian menunjukkan bahwa induk betina yang gonadnya berkembang mempunyai warna bintik hitam yang jelas, sebaliknya induk jantan warna bintik hitam memudar. Indeks kematangan gonad (IKG) yang diamati pada ikan uceng setelah 12 bulan pemeliharaan yaitu berkisar antara 0,007-0,027 pada jantan dan 0,13-0,25 pada betina. Kisaran diameter telur yang diamati yaitu berkisar antara 0,61-0,68 mm, dengan fekunditas 680-4.198 butir. Sedangkan pola pertumbuhannya menunjukkan bahwa ikan uceng betina dan jantan memiliki pola pertumbuhan allometrik negatif (b= 2,739 pada betina; b= 2,895 pada jantan). Nilai faktor kondisi Fulton (K) pada ikan uceng yang diamati yaitu 0,44-1,07 (rata-rata ± SD: 0,70 ± 0,11) pada betina dan 0,37-0,72 (rata-rata ± SD: 0,60 ± 0,06) pada jantan. Dari pengamatan ini ditemukan bahwa proses perkembangan kematangan gonad ikan uceng di akuarium lebih lambat dibandingkan ikan uceng di habitat aslinya.Barred loach (Nemacheilus fasciatus) is one of native fish species in Indonesia with high economic value, where their availability still depends on wild capture. Thus, domestication is needed to maintain its sustainability. The purpose of this study was to observe the reproductive biology and growth pattern of barred loach reared in aquarium (artificial environment). Fish were collected from Progo River, Temanggung, Central Java (total length of: 5.55 ± 0.53 cm; body weight: of 2.49 ± 0.24 g). The fish were reared for 12 months in aquarium (40 cm × 30 cm × 30 cm) with flowthrough system and equipped with aeration. The fish were fed with Tubifex until they accepted commercial feed. Then, the fish were fed twice a day at a ratio of 3% from the total fish biomass. The reproductive biology data were collected by measuring their total length, body weight, gonad weight, fecundity, egg diameter, and gonadosomatic index. Growth pattern were measured from length, weight, and survival. The results showed that matured female had shown black spot and male had inconsistent formation of black spots on their body. The gonadosomatic indexes ranged between 0.007-0.027 for male and ranged between 0,13-0,25 for female. The egg diameters ranged between 0.61-0.68 mm, with the fecundity of 680-4198 eggs. The result of observation on growth pattern showed that female and male had negative allometric growth (b= 2.739 for female, b= 2.895 for male). The value of Fulton condition factor (K) in the observed fish was 0.44-1.07 (mean ± SD: 0.70 ± 0.11) on female and 0.37-0.72 (mean ± SD: 0.60 ± 0.06) on male. From this study, it was found that gonad maturity development of barred loach reared in aquarium was slower than those in their natural habitat.

Abstract

Kanibalisme yang tinggi adalah salah satu permasalahan utama dalam budidaya lobster. Penggunaan selter individu pada budidaya pendederan lobster dimaksudkan untuk meniadakan kontak langsung antar lobster dalam media budidaya. Tujuan penelitian ini adalah untuk menganalisis kualitas air dan pengaruh penggunaan selter individu terhadap kinerja produksi pendederan lobster pasir Panulirus homarus dengan sistem resirkulasi. Lobster berukuran 51,29 ± 7,26 g/ekor dipelihara selama 60 hari dengan pemberian pakan 3%-4% sebanyak satu kali per hari. Penelitian dilakukan dengan empat perlakuan dan dua ulangan. Jenis perlakuan yang digunakan, yaitu selter pipa PVC sebagai kontrol (K), selter individu persegi (SI n), selter individu segitiga (SI ), dan selter individu tabung (SI l). Hasil penelitian menunjukkan bahwa kualitas air budidaya menggunakan sistem resirkulasi masih memenuhi syarat untuk budidaya lobster. Penggunaan selter individu berpengaruh positif terhadap respons sintasan, tetapi berpengaruh negatif terhadap pertumbuhan lobster.High cannibalism was an important issue in lobster rearing. The use of individual shelter in lobster rearing was intended to eliminate a direct contact between lobsters in a rearing media. The purpose of this study was to analyze water quality and the effect of individual shelter on the growth response of spiny lobster (Panulirus homarus) juvenile reared in an aquaculture media equipped with a recirculation system. Lobsters with an average size of 51.29 ± 7.26 g were reared for 60 days and fed with trash fish of 3%-4% of the biomass daily. This study used four treatments with two replications. The treatments consisted of a PVC pipe shelter as control (K), individual square shelter (SI n), individual triangle shelter(SI ), and individual tube shelter (SI l).The results of the study showed that the water quality in the aquaculture media was still within the suitable range for lobster rearing. The use of individual shelter had positive effects on the survival rate but had negative effects on the growth of spiny lobster juvenile.
